What is this line for?? It has a one-way valve in the hose.

I bought a powder coated plenum and the guy plugged the port off the plenum.
Should I re-tap a hole or is there a way to go around it?

That hose contains a check valve for the braking system. You will need to retain the check valve and connect it to a vacuum source somewhere. It doesn't need to be connected to the port on the plenum.
