Check the air pressure in all of the tires, engine oil and a visual on the rest of the vital fluids. In the trunk . . . A roll of paper towels, 1 bottle of detailing spray, 1 bottle of water, small container of hand cleaner and a pkg. of handy wipes. The items stored in the trunk will allow you to clean off bugs, road tar, etc. until you return and can wash your car.

Being somewhat of a road trip guru this is what I do:
1. Get the car serviced (especially if your trip will last longer than 1,000 miles round trip).
2. Depending on how much water you drink, buy a pack of water. I was happy enough with a six pack of Dasani bottles from York, PA back into the NY metro area (3 hours or so).
3. Pack in the snacks but go easy on sodium! Buy low sodium snack foods because I was so dried out from the beef jerky and Cheeze-its that my tongue was ready to die.
4. Re-stock your entire in-car cd collection and if you don't have satellite radio, pick up one of the receiver units from a company like Tao (about $130 for XM + subscription). I ended up not listening to my cd collection most of the time and was instead tuned into the variety of satellite radio. (Hopefully the new G will have an mp3 player hook up so we can use those too!)
5. Don't forget your in-car cell phone charger.
That's all I can think of for now... Use that with everything everyone else mentioned and you should be good.
Also, listen to Andy about the hand wipes.. eating all that stuff in the car and then getting it on your steering wheel etc.. it's better to have a cleaner on hand. Where you going btw
