You can and should do it whenever you want to. It removes contaminants that are stuck in the paint. Run your hand along the hood of your car. If it feels rough at all, clay it. After claying it will be super silky smooth because you removed all the stuff stuck in it. I wouldn't do it in temps under 50 degrees F because it may cause some marring due to the inability of the clay to completely soften. Others may disagree.

I think I am using the Meguirs kit, and they say the clay bar is good for about 3 cars.
Always much easier to cut it smaller and never have to think about it,
